The movie story deals with three friend Amarnath, Gopal and his sister, Mala. As Mala and Gopal's mom is a widow and is unable to care for both the children, she decides to let her Bombay-based cousin, Shobha R. Sharma, adopt Mala. Gopal is always in trouble with the villagers for petty theft, and when he learns that Mala has left, he too runs away from home. Years later, all three have grown up. Mala's mom is deceased, while she continues to live with her aunt and uncle, Public Prosecutor Ramnath Sharma; Amarnath is now an Advocate working with Senior Advocate Mehta, and is in love with Mala, not knowing that she is his childhood friend; while Gopal has been to prison in Poona, Madras, and the Police in Hyderabad want him for questioning. Soon Amarnath will be defending Gopal, alias Ram Singh, for the murder of one Mohanchand - and it will be an uphill task for Amarnath - for not only do the Police have airtight evidence against Gopal, Mohanchand was to be the future son-in-law of Ramnath - who has sworn to bring Gopal to justice by hook or by crook.

Pratap lives a wealthy lifestyle in a mansion along with his mother, Maharani. He is married to Parvati but the couple are childless. Then differences arise between the two on one hand, and Maharani and Pratap on the other. Parvati leaves and moves in with Dr. Bharati and his wife, while Pratap moves out to his own mansion. When Pratap gets the news that Parvati has given birth to a boy, he goes to visit her and secretly abducts the child. When he is unable to find a woman to breast-feed the child, he patches-up with Parvati and she moves back in along with an orphaned child, Shomu. The couple name their child, Shankar, who becomes friendly with Shomu much to the chagrin of Pratap, who instructs his staff to house Shomu in the stables. Parvati loves both boys equally, and the two children grow up almost like brothers, with Shankar falling in love with wealthy Seema, and Shomu wooing village belle Shaguna much to the displeasure of her to-be father-in-law, Balram, who complains to Pratap, who, in turn, asks Shomu to leave. Subsequently Shomu falls very ill and Parvati wants to go and see him. Pratap makes it clear that if she leaves - then she will not be welcomed back. The question remains what will Parvati do under these circumstances?

Dewan Bahadu Atal Rai believed that both his sons, Pavitra Kumar (alias Puttan) and Charitra Kumar (also known as Chuttan) were nothing better than loafers and ne'er-do-wells. Their mother Malati stubbornly pinned her faith in Chuttan and Puttan and asserted that they were still mere children. Let us find brides for them, and then watch the results! This public exhibition of their love enraged their father, who took the extreme step and turned them out of his house. Still hoping to win the hearts of their beloved, Chuttan and Puttan hatch a plot. Saying that he had murdered his brother Puttan, Chuttan rushed to Shaila to seek refuge. The poor girl believed his story, and tried to shield him from prying eyes. Meanwhile, Puttan took to a disguise, which led him to a smuggler's den. The criminals discovered him in their midst, and after knocking him unconscious, he was thrown into a convenient lake. Now Puttan really disappeared, and to his brother Chuttan's ill luck a body floated in the lake on which was found Puttan's identification in the shape of his jacket and watch. Chuttan was wanted for murder! The joke had boomeranged. Puttan's father believed that his son was still alive, a faith shared by Shaila. Puttan's photograph was advertised widely, with a reward offered to anyone reporting his whereabouts. Now enter into the drama Mani, who bore a striking resemblance to Puttan, his mother, and his bethrothed Rukmini. Two desperados notice the resemblance, and kidnapping the protesting Mani, carry him off to Bombay to claim the rich reward from Diwan Bahadur Atal Rai. Incidents follow, fast and furious, and the story of "Do Phool" races towards an unexpected climax, providing exhilarating entertainment as laughs and thrills loom as large as life. Such incidents are better enjoyed upon the screen, and it is left to the viewer to decide whether Chuttan and Puttan are `Do Phool' or really `Two Fools'.
